When asked if the mission structure in “GTA 5” is similar to the one found in “GTA IV: Episodes from Liberty City,” Matt Bertz (the Managing Editor at Game Informer), responded with the following comment on Twitter:

No, they said it's not like episodes. Some story missions are just one guy. Some a couple, some have all three.

When asked if you can use one character for a mission and then play as another one in the second play-through, responded with the following:
Not for individual missions.

But within missions with multiple characters, you can switch between them on the fly.

I've read this in other articles/threads too. Rockstar wants people to play ALL the characters storylines, to not just pick one character, and play the game through with that character. So as described above, you can switch between characters stories, which have their own individual missions, and when you play a mission that has multiple characters you can switch between them on the fly. Leslie Benzies, one of the producers had this to say in an IGN article:

"One character might have an ambient activity that takes them out in to the countryside; if the player decides that they want to do something different they can switch to another character and be back in the city ready to start their next mission," Benzies said. "Having three characters allows players to explore the whole map without having to worry about the long drive back to their next objective."

There's another article IGN did that I just put the icing on the cake (this is in relation to multiple character missions):

"Players will experience missions as different combinations of the three characters, and each will have a different skillset. For example, in one heist mission, Michael rappels down the side of a building while Franklin uses a sniper rifle from afar and Trevor flies in a helicopter above. Players can switch back and forth between Michael grabbing a target inside the building or Franklin trying to snipe enemies inside. Once you escape, the player can choose to fly the getaway helicopter as Trevor, snipe other pilots as Franklin or fire a gun from the back of the vehicle as Michael."
