On the Rain-Slick Precipice of Darkness will introduce those who haven't to the comic. While it's certainly true that you can play the game without any prior knowledge of what Penny Arcade is all about -- after all, the setting is completely different, and the characters are fighting evil things instead of remarking on videogames -- it's actually quite difficult to explain the world of New Arcadia outside the context of Penny Arcade.
